Claire Fitzsimmons, 51, booked £265 flights to Corfu for her and husband Nicks 25th wedding anniversary in September.
But they have been forced to scrap the trip after Nick, who suffers from a hereditary condition, had his left leg amputated below the knee.
Claire, a bank cashier, said: “My husband suffers from a condition which causes him to produce too much cholesterol. His arteries in his legs blocked up and doctors were left with no choice but to amputate.
“It was devastating. It does say on the Ryanair website that in cases of serious illness and death, refunds would be given.
“I emailed them and received a standard reply, saying no to a refund. I was absolutely disgusted.”

In a letter, Claire was told: “I sympathise with your circumstances, but Im afraid I cannot accede to your request for a refund in this case.”
The wife added: “Weve been through a lot already and this just made everything harder.” Ryanair had last night failed to comment on the couples claims.
Meanwhile, more than 150 Ryanair flights serving London Stansted were cancelled or delayed yesterday due to bad weather. Passengers reported long queues and a lack of information.
A spokesman for the airline said: “Ryanair sincerely apologise for these weather and air traffic control disruptions, which are beyond our control.”
